• DES, also called desmin, is a protein that in humans is encoded by the DES gene, and this gene is mapped to 2q35.
• DES is the muscle-specific member of the intermediate filament (IF) protein family.
• It is one of the earliest myogenic markers, both in heart and somites, and is expressed in satellite stem cells and replicating myoblasts.
• DES is very important in muscle cell architecture and structure since it connects many components of the cytoplasm.
• It may be also play an important role in mitochondria function.
• DES provides attachments between the terminal Z disc and membrane-associated proteins to form a force-transmitting system that parallels the thin filaments at myotendinous junctions.
• Applications:Suitable for use in Western Blot and Immunohistochemistry.
• Other applications not tested.
• Recommended Dilution:Western Blot: 0.1-0.5ug/ml, the detection limit is ~0.25ng/lane under reducing conditions
• Immunohistochemistry (frozen): 0.5-1ug/mlImmunohistochemistry (paraffin): 0.5-1ug/ml, boiling the paraffin sections in 10mM citrate buffer, pH 6.0, for 20 mins is required for the staining of formalin/paraffin sections
• Optimal dilutions to be determined by the researcher.
• Storage and Stability:Lyophilized powder may be stored at -20°C.
• Stable for 12 months after receipt at -20°C.
• Reconstitute with sterile ddH2O.
• Aliquot to avoid repeated freezing and thawing.
• Store at -20°C.
• Reconstituted product is stable for 12 months at -20°C.
• For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
• Further dilutions can be made in assay buffer.
