CD43, also known as leukosialin or sialophorin, is a transmembrane cell surface protein that in humans is encoded by the SPN gene.
It is mapped to 16p11.2.
It is a major sialoglycoprotein on the surface of human T lymphocytes, monocytes, granulocytes, and some B lymphocytes, which is important for immune function and may be part of a physiologic ligand-receptor complex involved in T-cell activation.
Expression of CD43 is deficient and/or defective in the X-chromosome-linked immunodeficiency disorder Wiscott-Aldrich syndrome, suggesting that CD43 have a role in T-cell activation.
T-cell activation requires the removal of CD43 from the immunologic synapse to allow efficient engagement of the TCR with molecules on the APC.
Applications:Suitable for use in Western Blot, Immunohistochemistry and Immunocytochemistry.
Other applications not tested.
Recommended Dilution:Western Blot: 0.1-0.5ug/ml, the detection limit is ~0.25ng/lane under reducing conditions
Immunohistochemistry (paraffin): 0.5-1ug/ml, boiling the paraffin sections in 10mM citrate buffer, pH 6.0, for 20 mins is required for the staining of formalin/paraffin sections
Immunocytochemistry: 0.5-1ug/mlOptimal dilutions to be determined by the researcher.
Storage and Stability:Lyophilized powder may be stored at -20°C.
Stable for 12 months after receipt at -20°C.
Reconstitute with sterile ddH2O.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Reconstituted product is stable for 12 months at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
