Protein unc-13 homolog D (Munc13-4) is involved in cytotoxic granule exocytosis in lymphocytes.
It regulates assembly of recycling and late endosomal structures which leads to the formation of an endosomal exocytic compartment.
It also regulates Ca2+-dependent secretory lysosome exocytosis in mast cells.
Munc13- 4 is highly expressed in the spleen, thymus and leukocytes and to a lesser extent in the lung and placenta, brain, heart, skeletal muscle and kidney.
Defects in Munc13-4 are the cause of hemophagocytic lymphohistiocytosis familial type 3 (FHL3).
