Presenilin enhancer protein 2 (PEN), a 12kD member of the PEN2 family and subunit of the gamma-secretase complex.
This complex is an endoprotease that cleaves integral membrane proteins.
These include Notch receptors and the beta-amyloid precursor protein which is cleaved to form amyloid beta peptide, a major component of the neuritic plaques found in Alzheimer’s disease.
The gamma secretase complex consists of nicastrin, the presenilin homodimers and APH1.
It is the addition of PEN2, the final stage of gamma secretase maturation that facilitates endoproteolysis of presenilin and confers proteolytic activity.
PEN2 expression has been observed in leukocytes, lung, placenta, small intestine, liver, kidney, spleen thymus, skeletal muscle, heart and brain.
