XIAP is an endogenous inhibitor of cell death, which functions by suppressing caspases 3, 7, and 9.
The protein is also reported to play a a role in the activation of NF kappa B and the regulation of cyclins and cyclin-dependent kinase inhibitors (CDKIs).
Regulators of XIAP activity include XAF-1 and Smac.
Applications:Suitable for use in Western Blot and Immunohistochemistry.
Other applications not tested.
Recommended Dilution:Western Blot: detects a band of ~57kD in human kidney lysates
Immunohistochemistry (paraffin): Requires antigen retrieval using heat treatment prior to staining of paraffin sections
Optimal dilution determined by the researcher.
Recommended Secondary Reagents:Sheep anti rabbit IgG: HRP
Storage and Stability:Store product at 4°C if to be used immediately within two weeks.
For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C.
Aliquots are stable at -20°C for 12 months after receipt.
Dilute required amount only prior to immediate use.
Further dilutions can be made in assay buffer.
Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Note: Applications are based on unconjugated antibody.
