PUMA, also known as bbc3, is a pro-apoptotic member of the BCL-2 family.
PUMA is localized to the mitochondria, binds BCL-2 and induces cytochrome C release and apoptosis in response to p53.
Applications:Suitable for use in Immunohistochemistry and Western Blot.
Other applictions not tested.
Recommended Dilutions:Western Blot: 2-4ug/ml.
Detects a band at ~23kD in K562 cell lysates
Immunohistochemistry (paraffin): 10ug/ml.
Requires antigen retrieval using heat treatment prior to staining of paraffin sections.
Sodium citrate buffer pH 6.0 is recommended
Optimal working dilutions to be determined by researcher.

Recommended Secondary Reagents:Sheep anti Rabbit IgG (HRP)
Storage and Stability:Store product at 4°C if to be used immediately within two weeks.
For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C.
Aliquots are stable at -20°C for 12 months after receipt.
Dilute required amount only prior to immediate use.
Further dilutions can be made in assay buffer.
Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Note: Applications are based on unconjugated antibody.
