MaxLight™750 is a new Near IR stable dye conjugate comparable to DyLight™750, Alexa Fluor™700 and offers better labeling efficiency, brighter imaging and increased immunodetection.
Type 2 protein phosphatases are serine/threonine phosphatases which can preferentially dephosphorylate the alpha subunit of phosphorylase kinase and are insensitive to inhibitor 2 treatment.
Protein phosphatase 2A (PP2A) can be distinguished from PP2B and PP2C which require Ca2+ and Mg2+, respectively, by its partial activity in the absence of divalent cations.
PP2A activity is potently inhibited by treatment with okadaic acid and shows much stronger activity toward histone phosphorylated by Protein Kinase C and casein phosphorylated by Protein Kinase A than other protein phosphatases.
Applications:Suitable for use in Western Blot, Immunoprecipitation and Phosphatase assays.
Other applications have not been tested.
Recommended Dilutions:Western Blot: Detects PP2A catalytic subunit (~34kD) in rabbit brain membrane cytosol and in human A431 RIPA cell lysate.
Immunoprecipitation: immunoprecipitates PP2A catalytic subunit from 500ug rabbit brain extract and 1mg human A431 RIPA cell lysate
Immunoprecipitation/Phosphatase Assay: 5ug immunoprecipitates PP2A from 1U of purified Protein Phosphatase 2A, Serine, Threonine (PP2A Ser/Thr).
Phospatase activity was demonstrated using p-Nitrophenyl Phosphate (pNPP, KR-pT-IRR), Ser/Thr Phosphatase Assay Kit, BioAssay™ (Catalog No: N2600-35)
Optimal dilutions to be detemined by the researcher.
Storage and Stability:Store product at 4°C in the dark.
DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid.
Dilute required amount only prior to immediate use.
Further dilutions can be made in assay buffer.
Caution: MaxLight™750 conjugates are sensitive to light.
For maximum recovery of product, centrifuge the original vial prior to removing the cap.
Note: Applications are based on unconjugated antibody.
