MaxLight™650 is a new Far-IR stable dye conjugate comparable to Alexa Fluor™647, DyLight™649, Cy5™ and offers better labeling efficiency, brighter imaging and increased immunodetection.
Pin1 is a peptidyl-prolyl isomerase which is present throughout the cell cycle and which is thought to alter its mitotic targets by changing their conformation.
Many of the substrates of the mitotic regulator PIN1 are recognized by the monoclonal antibody, clone MPM-2.
This clone recognizes a subset of M phase phosphoproteins in a phosphorylation-dependent manner.
It has also been shown that RNA polyermase II large subunit interacts with PIN1 in HeLa cells and may thus regulate transcriptional and post-transcriptional events by catalyzing phosphorylation-dependent conformational changes.
Applications:Suitable for use in Western Blot and Immunoprecipitation.
Other applications not tested.
Recommended Dilutions:Western Blot: 0.5-2ug/ul.
Detected Pin1 in RIPA lysates from rat PC-12 and human HeLa cells
Immunoprecipitation: 4ug
Optimal dilutions to be determined by the researcher.
Positive Antigen Control Included:F4209: Fibroblast Cell, 3T3/A31.
Add 2.5ul of 2-mercaptoethanol for each 100ul of lysate and boil for 5 minutes to reduce the preparation.
Load 20ul of reduced lysate per lane for minigels.
Storage and Stability:Store product at 4°C in the dark.
DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid.
Dilute required amount only prior to immediate use.
Further dilutions can be made in assay buffer.
Caution: MaxLight™650 conjugates are sensitive to light.
For maximum recovery of product, centrifuge the original vial prior to removing the cap.
Note: Applications are based on unconjugated antibody.
