MaxLight™650 is a new Far-IR stable dye conjugate comparable to Alexa Fluor™647, DyLight™649, Cy5™ and offers better labeling efficiency, brighter imaging and increased immunodetection.
Histone H3 is one of the five main histone proteins involved in the structure of chromatin in eukaryotic cells.
Featuring a main globular domain and a long N-terminal tail, H3 is involved with the structure of the nucleosomes of the 'beads on a string' structure.
The N-terminal tail of histone H3 protrudes from the globular nucleosome core and can undergo several different types of epigenetic modifications that influence cellular processes.
These modifications include the covalent attachment of methyl or acetyl groups to lysine and arginine amino acids and the phosphorylation of serine or threonine.
Applications:Suitable for use in FLISA, Western Blot and Immunocytochemistry.
Other applications not tested.
Recommended Dilutions:FLISA: ≥1:2000 is specific for peptides containing dimethylated Arg17.
No crossreactivity detected with a peptide containing dimethylated Arg26
Western Blot: 1:1000-1:2000 detected in-vitro methylated Histone H3.
Products from an in vitro methyl transferase reaction were resolved by electrophoresis and probed with H5110-14G.
Proteins were visualized using an IgG (HRP) Gt xRb* and a chemiluminescence detection system
Immunocytochemistry: Reported to show positive immunostaining for CARM-1 specific methylation of Histone H3 in 3134 cells.
Optimal dilutions to be determined by the researcher.

Storage and Stability:Store product at 4°C in the dark.
DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid.
Dilute required amount only prior to immediate use.
Further dilutions can be made in assay buffer.
Caution: MaxLight™650 conjugates are sensitive to light.
For maximum recovery of product, centrifuge the original vial prior to removing the cap.
Note: Applications are based on unconjugated antibody.
