MaxLight™490 is a new Blue-Green photostable dye conjugate comparable to DyLight™488, Alexa Fluor™488 and offers better labeling efficiency, brighter imaging and increased immunodetection.
ADAM10 is a member of the ADAM (a disintegrin and metalloprotease like domain) family.
It cleaves the membrane-bound precursor of TNF-alpha at 76-Ala-|-Val-77 to its mature soluble form.
It is responsible for the proteolytic release of several cell-surface proteins, including heparin binding epidermal growth-like factor, ephrin-A2 and for constitutive and regulated alpha-secretase cleavage of amyloid precursor protein (APP).
It contributes to the normal cleavage of the cellular prion protein and is involved in the cleavage of the adhesion molecule L1 at the cell surface and in released membrane vesicules, suggesting a vesicle-based protease activity.
Applications:Suitable for use in Western Blot, Immunohistochemistry and Immunofluorescence.
Other applications have not been tested.
Recommended Dilutions:Western Blot: 1:500-1:2000 detected ADAM 10 on 10ug of RAW264.7 lysates.
RAW264.7 lysate was resolved by electrophoresis, transferred to PVDF membrane and probed with A0859-20 (1:1000).
Proteins were visualized using an IgG (HRP) donkey anti-rabbit and chemiluminesence detection system.
Immunohistochemistry: A previous lot of this antibody was used to detect ADAM10 in formalin-fixed, paraffin-embedded renal tissue and renal cell carcinoma tissue (Gutwein, P., et al., 2009)
Immunofluorescence: A previous lot of this antibody was used to detect ADAM10 in RCC4 and A498 cells (Gutwein, P., et al., 2009)
Optimal dilutions to be determined by the researcher.
Storage and Stability:Store product at 4°C in the dark.
DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid.
Dilute required amount only prior to immediate use.
Further dilutions can be made in assay buffer.
Caution: MaxLight™490 conjugates are sensitive to light.
For maximum recovery of product, centrifuge the original vial prior to removing the cap.
Note: Applications are based on unconjugated antibody.
