MaxLight™490 is a new Blue-Green photostable dye conjugate comparable to DyLight™488, Alexa Fluor™488 and offers better labeling efficiency, brighter imaging and increased immunodetection.
The catalytic domain of serum and glucocorticoid-regulated protein kinase (SGK) is 54% identical with that of Akt/PKB.
Although SGK1 lacks the PtdIns(3,4,5) P3-binding Pleckstrin-homology domain, it retains the residues that are phosphorylated by PDK1 (Thr256 and Ser422).
PDK1 activates SGK in vitro by phosphorylating Thr256.
Like Akt/PKB, SGK1 preferentially phosphorylates serine and threonine residues that lie in RXRXXS/T motifs.
SGK1 can be activated by PI-3 Kinase, which makes it an important signaling molecule in growth factor and insulin dependent signaling pathways.
Applications:Suitable for use in Western Blot and Immunoprecipitation.
Other applications have not been tested.
Recommended Dilution:Western Blot Analysis: 0.5-2ug/ml detects SGK1 in RIPA lysates from A431 and Rat 2 cells.
Rat 2 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with S1010-83 (0.5ug/ml).
Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system
Immunoprecipitation: 4ug Immunoprecipitates SGK1 from 500ug of Rat 2 lysate
Optimal dilutions to be determined by the researcher.
Storage and Stability:Store product at 4°C in the dark.
DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid.
Dilute required amount only prior to immediate use.
Further dilutions can be made in assay buffer.
Caution: MaxLight™490 conjugates are sensitive to light.
For maximum recovery of product, centrifuge the original vial prior to removing the cap.
Note: Applications are based on unconjugated antibody.
