MaxLight™405 is a new Violet photostable dye conjugate comparable to Alexa Fluor® 405, PacificBlue™, Brilliant™ Violet 421 and offers better labeling efficiency, brighter imaging and increased immunodetection.
AS160 (Akt Substrate of 160kD, TBC1D4) is a Rab-GAP protein that regulates the trafficking of GLUT4 (glucose transporter 4) to the plasma membrane.
AS160 is known to be phosphorylated on at least 8 residues following stimulation from IGF-1, EGF, PMA, and AICAR (Chen, S., 2008).
Thr642 is phosphorylated by Akt in response to IGF-1, while Ser341 and Thr642 of AS160 are phosphorylated by RSK (p90 ribosomal S6 kinase) in response to PMA.
Ser588 has been shown to be phosphorylated by AMPK in vitro, though other kinases might be responsible as well.
The phosphorylation site, Ser666, does not contain the Akt consensus sequence.
Its phosphorylation is suppressed by AICAR and by combined mutations of TOS (mTOR signaling)-like sequence (FEMDI) and rapamycin (Geraghty, K.M., 2007).
Additionally, its phosphorylation is markedly increased in response to IGF-1 and this phosphorylation is blocked in response to the PI3 Kinase inhibitor LY294002 (Geraghty, K.M., 2007).
Applications:Suitable for use in Western Blot.

Storage and Stability:Store product at 4°C in the dark.
DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid.
Dilute required amount only prior to immediate use.
Further dilutions can be made in assay buffer.
Caution: MaxLight™405 conjugates are sensitive to light.
For maximum recovery of product, centrifuge the original vial prior to removing the cap.
Note: Applications are based on unconjugated antibody.
