MaxLight™405 is a new Violet photostable dye conjugate comparable to Alexa Fluor® 405, PacificBlue™, Brilliant™ Violet 421 and offers better labeling efficiency, brighter imaging and increased immunodetection.
KCC2 is a member of the cation-chloride cotransporter gene family.
It acts as a K-Cl cotransporters.
KCCs normally lower intracellular chloride concentrations below the electrochemical equilibrium potential and depending on the chemical concentration gradients of potassium and chloride, KCC2 can operate as a net efflux or influx pathway.
It is proposed to act as the main chloride extruder to promote fast hyperpolarizing postsynaptic inhibition in the brain.
KCC2 is expressed at high levels in neurons throughout the nervous system and immunofluorescence shows that the protein is localized at inhibitory synapses of the spinal cord.
Studies in mice have shown that KCC2 reduces GABA's inhibitory signaling, resulting in motor defects, epilepsy, and anxiety-like behavior.
Applications:Suitable for use in Western Blot and Immunocytochemistry.
Other applications not tested.
Recommended Dilutions:Western Blot: 1ug/ml detected KCC2 in rat brain membrane preparations.
Rat brain membrane preparation were resolved by electrophoresis, transferred to nitrocellulose and probed with K0120-07.
Proteins were visualized using an IgG (HRP) goat anti-rabbit and a chemiluminescence detection system.
Immunocytochemistry: 1:200 detected KCC2 in MDCK cells as reported by an independent laboratory
Optimal dilutions to be determined by the researcher.
Storage and Stability:Store product at 4°C in the dark.
DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid.
Dilute required amount only prior to immediate use.
Further dilutions can be made in assay buffer.
Caution: MaxLight™405 conjugates are sensitive to light.
For maximum recovery of product, centrifuge the original vial prior to removing the cap.
Note: Applications are based on unconjugated antibody.
