MaxLight™405 is a new Violet photostable dye conjugate comparable to Alexa Fluor® 405, PacificBlue™, Brilliant™ Violet 421 and offers better labeling efficiency, brighter imaging and increased immunodetection.
Histone H3 is one of the five main histone proteins involved in the structure of chromatin in eukaryotic cells.
Featuring a main globular domain and a long N-terminal tail, H3 is involved with the structure of the nucleosomes of the “beads on a string” structure.
The N-terminal tail of histone H3 protrudes from the globular nucleosome core and can undergo several different types of epigenetic modifications that influence cellular processes.
These modifications include the covalent attachment of methyl or acetyl groups to lysine and arginine amino acids and the phosphorylation of serine or threonine.
Applications:Suitable for use in Western Blot, Dot Blot, Peptide Inhibition Assay and Immunofluorescence.
Other applications not tested.
Recommended Dilution:Western Blot: 1:500 of this lot detected trimethyl Histone H3 in acid extracted proteins from HeLa cells
Peptide Inhibition: Specificity of this lot confirmed by the ability of 10uM of the immunizing peptide to completely abolish detection of histone H3 in Western Blot of HeLa acid extracts.
No inhibition of detection was observed by preabsorption of the antibody with 10uM histone H3 peptide containing mono- or dimethyl-lysine 9, or mono-, di- or trimethyl-lysine 27 modifications

Storage and Stability:Store product at 4°C in the dark.
DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid.
Dilute required amount only prior to immediate use.
Further dilutions can be made in assay buffer.
Caution: MaxLight™405 conjugates are sensitive to light.
For maximum recovery of product, centrifuge the original vial prior to removing the cap.
Note: Applications are based on unconjugated antibody.
