• MaxLight™405 is a new Violet photostable dye conjugate comparable to Alexa Fluor® 405, PacificBlue™, Brilliant™ Violet 421 and offers better labeling efficiency, brighter imaging and increased immunodetection.
• Absorbance (400nm); Emission (423nm); Extinction Coefficient 32,000.
• Lat is an adapter protein expressed in lymphoid cells, which is a substrate for ZAP70 and Syk protein kinases.
• Phosphorylated LAT associates with GRB2, PLC-gamma1 and P85 PI 3-K.
• Expression of a mutant LAT protein missing tyrosine phosphorylation sites leads to inhibition of T-cell activation, suggesting that the protein is an important signal transducer in T-cells
• Applications:Suitable for use in Western Blot.
• Other applications have not been tested.
• Recommended Dilution:Western Blot: 0.5-2ug/ml detects phospho-LAT (Tyr191) in RIPA lysates from Jurkat cells stimulated with C305.
• Jurkat cells were incubated without or with, a monoclonal antibody that binds Jurkat T-cell receptors.
• Cell lysates were prepared, resolved by electrophoresis, transferred to nitrocellulose and probed with anti-phospho-LAT (Tyr191) (2ug/ml), or anti- LAT (1ug/ml).
• Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system
• Optimal dilutions to be determined by the researcher.
• Storage and Stability:Store product at 4°C in the dark.
• DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid.
• Dilute required amount only prior to immediate use.
• Further dilutions can be made in assay buffer.
• Caution: MaxLight™405 conjugates are sensitive to light.
• For maximum recovery of product, centrifuge the original vial prior to removing the cap.
• Note: Applications are based on unconjugated antibody.
