Histone H3 is one of the five main histone proteins involved in the structure of chromatin in eukaryotic cells.
Featuring a main globular domain and a long N-terminal tail, H3 is involved with the structure of the nucleosomes of the 'beads on a string' structure.
The N-terminal tail of histone H3 protrudes from the globular nucleosome core and can undergo several different types of epigenetic modifications that influence cellular processes.
These modifications include the covalent attachment of methyl or acetyl groups to lysine and arginine amino acids and the phosphorylation of serine or threonine.
Histone variant H3.3 is typically enriched in active chromatin.
Applications:Suitable for use in Western Blot, Chromatin Immunoprecipitation, Dot Blot and Immunocytochemistry.
Other applications not tested.
Recommended Dilution:Optimal dilutions to be determined by the researcher.
Positive Control:HeLa acid extract
Storage and Stability:Store product at 4°C if to be used immediately within two weeks.
For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C.
Aliquots are stable at -20°C for 12 months after receipt.
Dilute required amount only prior to immediate use.
Further dilutions can be made in assay buffer.
Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Note: Applications are based on unconjugated antibody.
