Rsk1, Rsk2, and Rsk3 (ribosomal protein S6 kinase, 90kDa, polypeptide 1,2,3; S6K-alpha 1,2,3) are Ser/Thr kinases which are activated by MAP Kinases, and are alternatively referred to as MAPKAP kinases 1a, 1b, and 1c.
Following activation, Rsk isoforms translocate to the nucleus, where Rsk2 phosphorylates Ser10 of Histone H3, a critical step in the stimulation of immediate-early gene expression.
Rsk1 is able to phosphorylate Ser133 of CREB, activating it as a transcription factor.
Rsk1 activation can be monitored by phosphorylation state specific antibodies directed against sites involved in its activation.
Applications:Suitable for use in ELISA, Western Blot and Immunoprecipitation.
Other applications not tested.
Recommended Dilution:Optimal dilutions to be determined by the researcher.
Storage and Stability:Store product at 4°C if to be used immediately within two weeks.
For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C.
Aliquots are stable at -20°C for 12 months after receipt.
Dilute required amount only prior to immediate use.
Further dilutions can be made in assay buffer.
Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Note: Applications are based on unconjugated antibody.
