Vav proteins belong to the guanidine nucleotide exchange factor family of proteins that couple receptors to Rho-GTPases.
The VAV family has currently three members in mammalian cells (VAV 1, VAV 2, and VAV 3).
VAV 1 contains multiple protein binding motifs, indicating a role for VAV 1 as an adaptor protein linking cell surface receptors to downstream signaling proteins.
VAV 1 is exclusively expressed in hematopoietic cells and plays a significant role in lymphocyte development and antigen receptor mediated activation of NFAT and NF(kappa)B, and in more delayed responses such as the production of cytokines, including IL-2, IL-3, IFN lambda, and TNF alpha.
VAV 3 is also involved in activating JNK, p21-activated kinase (PAK) and PLC lambda, implicating a critical role in cytoskeletal rearrangement.
VAV 3 is inducibly tyrosine phosphorylated in response to stimulation of the epidermal growth factor (EGFR), platelet-derived growth factor (PDGFR) and T-cell receptors (TCR), enhancing its guanine nucleotide exchange factor activity in vitro.
Tyrosine 173 is the regulatory phosphorylation site of VAV 3.
Applications:Suitable for use in Western Blot, Immunoprecipitation, Immunocytochemistry and Immunohistochemistry.
Other applications not tested.
Recommended Dilution:Western Blot: detected Vav3 in RIPA lysates from HeLa and Jurkat cells
Immunoprecipitation: Reported to immunoprecipitate Vav3 from Jurkat cell lysate and SU86.86 pancreatic tumor cell lysate
Immunocytochemistry: Reported to detect Vav3 in Jurkat and SU86.86 pancreatic tumor cells
Immunohistochemistry: Reported to detect Vav3 in pancreatic adenocarcinoma sections.
Optimal dilutions to be determined by the researcher.
Storage and Stability:Store product at 4°C if to be used immediately within two weeks.
For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C.
Aliquots are stable at -20°C for 12 months after receipt.
Dilute required amount only prior to immediate use.
Further dilutions can be made in assay buffer.
Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Note: Applications are based on unconjugated antibody.
