Applications:Suitable for use in Western Blot and Immunoprecipitation.
Other applications have not been tested.
Recommended Dilution:Western Blot: 0.5-2.0ug/ml detects MEK1 in RIPA lysates from A431 and 3T3/A31 cells.
A431 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with M2865-07 (0.5ug/ml).
Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system.
MEK1 (~45kD).
Immunoprecipitation: 4ug immunoprecipitates MEK1 from 500ug of A431 and 3T3/A31 RIPA lysate.
MEK1 Immunoprecipitation Kinase Assay, nonradioactive:4ug of this lot immunoprecipitated active MEK1 from NGF stimulated PC-12 cell lysates, as well as active human MEK1
Optimal dilutions to be determined by the researcher.
Add 2.5ul of 2-mercaptoethanol/100ul of lysate and boil for 5 minutes to reduce the preparation.
Load 20ug of reduced lysate per lane for minigels.
Storage and Stability:Store product at 4°C if to be used immediately within two weeks.
For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C.
Aliquots are stable at -20°C for 12 months after receipt.
Dilute required amount only prior to immediate use.
Further dilutions can be made in assay buffer.
Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Note: Applications are based on unconjugated antibody.
