MAD1 is a non-essential gene that encodes a component of the spindle checkpoint.
The spindle checkpoint delays the onset of anaphase in cells with defects in mitotic spindle assembly or in the attachment of chromosomes to the spindle microtubules.
The checkpoint works by inhibiting the activity of the anaphase promoting complex, thereby preventing the degradation of several cell cycle regulators.
Like other spindle checkpoint mutants, MAD1 loss-of-function mutants are sensitive to benomyl and cannot delay cell division in response to spindle depolymerization.
Mad1p becomes hyperphosphorylated upon spindle depolymerization.
Applications:Suitable for use in Immunofluorescence, Western Blot, Immunocytochemistry.
Other applications not tested.
Recommended Dilution:Western Blot: 1:100.
Detects a band of ~83kD Optimal dilutions to be determined by the researcher.
Storage and Stability:May be stored at 4°C for short-term only.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Aliquots are stable for 12 months after receipt.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
