• Recognizes an epitope within the N-Terminal (NT) region of both MAP1LC3A (Microtubule-associated proteins 1A/1B light chain 3A/LC3A) and MAP1LC3B (Microtubule-associated proteins 1A/1B light chain 3B/LC3B), ubiquitin-like proteins and members of the MAP1LC3 family, which are widely used as reliable markers for the monitoring of autophagy.
• LC3-I is the cytosolic form of LC3, which is converted into the active, membrane-bound form LC3-II, during the autophagy process.
• Tracking the level of conversion of LC3-I to LC3-II provides an indicator of autophagic activity, and levels of LC3-II in particular, correlate with the extent of autophagosome formation, due to its association with the autophagosome membrane.
• Applications:Suitable for use in Immunofluorescence and Western Blot.
• Other applications not tested.
• Recommended Dilution:Western Blot: 1:100-1:250, detects a band of ~14-15kD corresponding to LC3-II, and a band of ~17kD corresponding to LC3-I, in HeLa cell lysates
• Optimal dilutions to be determined by the researcher.
• Storage and Stability:May be stored at 4°C for short-term only.
• Aliquot to avoid repeated freezing and thawing.
• Store at -20°C.
• Aliquots are stable for 12 months after receipt.
• For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
