AIF1, otherwise known as IBA1, an actinbinding protein first identified in rat cardiac allografts with chronic rejection, which has been shown to enhance membrane ruffling, actin bundling activity of LCP1 (lymphocyte cytosolic protein 1) and RAC activation.
Has been used successfully for the identification of AIF1 expressing cells in an inflammatory model in formalin fixed mouse brain following heat mediated antigen retrieval with sodium citrate buffer by immunohistochemistry (Wang et al.
2014).
Applications:Suitable for use in ELISA, Western Blot and Immunohistochemistry.
Other applications not tested.
Recommended Dilutions:ELISA: 1:128,000
Western Blot: 0.1-1ug/ml, ~16-17kD band observed in Human Frontal Cortex, Rat Brain and in Mouse Brain and lymph node lysates (calculated MW of 16.7kD according to Human NP_001614.3, 16.9kD according to Mouse NP_062340.1 and 16.8kD according to Rat NP_058892.1).
Primary incubation 1 hour at room temperature
Immunohistochemistry (Paraffin): 4-6ug/ml.
Requires heat-mediated antigen retrieval prior to staining paraffin sections.
Tris/EDTA buffer pH9.0 is recommended for this purpose
Optimal dilutions to be determined by the researcher.
Storage and Stability:May be stored at 4°C for short-term only.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Aliquots are stable for 12 months after receipt.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
