Alpha-fetoprotein (AFP) is a 65kD glycoprotein found in the serum of the mammalian fetal liver, yolk sac, and GI tract.
While AFP expression in adult cells is low, it is aberrantly expressed in adult liver cancer cells (1,2).
The tumor suppressor gene p53 and beta-catenin are both involved in the regulation of AFP expression.
In normal adult cells, p53 binds to the repressor region of the AFP gene, thereby blocking transcription.
Mutations in both p53 and beta-catenin are associated with aberrant expression of AFP.
Elevated serum levels are predictive of hepatocellular carcinoma (3).
Applications:Suitable for use in Immunofluorescence, Western Blot, Immunohistochemistry.
Other applications not tested.
Recommended Dilution:Western Blot: 1:500-1:2000Immunohistochemistry: 1:50-1:200Immunofluorescence: 1:10-1:100Optimal dilutions to be determined by the researcher.
Storage and Stability:May be stored at 4°C for short-term only.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Aliquots are stable for 12 months after receipt.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
