Tumor Necrosis Factor alpha (TNF-alpha) is a 17.3kD multi-functional pro-inflammatory cytokine primarily secreted by macrophages, but also produced by T and B lymphocytes, activated monocytes and fibroblasts.
TNF-alpha exists in both a transmembrane and mature soluble form and has many biological properties, including activation of transcription factor NF-KappaB, the induction of apoptosis in various tumor cell lines and the stimulation of cell differentiation and proliferation under certain conditions.
Studies have revealed a role for TNF-alpha in inflammation, septic shock, autoimmune disease and rheumatoid arthritis.
Applications: Suitable for use in neutralization, ELISA, and Western Blot.
Other applications have not been tested.
Recommended Dilution:Neutralization: 0.012-0.014ug/ml required to yield one-half maximal inhibition [ND50] of hTNF-a (0.25ng/ml).
ELISA: 0.2-0.4 ug/ml (using 100ul/well antibody solution) Used in conjunction with compatible secondary reagents, allows the detection of at least 0.2ng/well of recombinant hTNF-a.
Western Blot: 0.1-0.2ug/ml.
Used in conjunction with compatible secondary reagents the detection limit for recombinant hTNF-a is 1.5-3.0ng/lane, under either reducing or non-reducing conditions
Optimal dilutions to be determined by the researcher.
Storage and Stability:Store product at 4°C in the dark.
DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid.
Dilute required amount only prior to immediate use.
Further dilutions can be made in assay buffer.
Caution: PE conjugates are sensitive to light.
For maximum recovery of product, centrifuge the original vial prior to removing the cap.
Note: Applications are based on unconjugated antibody.
