As a member of B7 family, B7-H4 (VTCN1) plays a critical role in the regulation of antigen-specific immune responses.
B7-H4 is a co-stimulatory protein expressed in activated T cells and is involved in the negative regulation of cell-mediated immunity in peripheral tissues.
B7-H4 ligation to its receptor BTLA T-lymphocytes results in inhibition of T-cell activation, cytokine secretion, and the development of cytotoxicity.
B7-H4 protein is typically expressed on the surface of activated T-cells, B-cells, monocytes, and dendritic cells, but not in most normal tissues.
In tissues, B7-H4 is over expressed in breast, endometrial, ovarian, renal cell (RCC), and non-small cell lung carcinomas (NSCLC).
B7-H4 has been identified as an important cancer marker for breast and ovarian cancers.
