MEKK8, known as COT for cancer Osaka thyroid, is a MAP3K type protein kinase that was identified by its oncogenic transforming activity in cells.
COT activates the JNK cascade by phosphorylating MKK4/MAP2K4 and the ERK cascade by phosphorylating MEK1/MAP2K1.
COT also has been implicated in the induction of NF-kappaB-dependent transcription and the production of TNF-alpha and interleukin-2 during T lymphocyte activation.
At least two protein isoforms which result from the use of alternative translation initiation sites and differ in their N-terminus have been reported.
In the COT oncoprotein, the C-terminal 70aa of normal COT are replaced by 18 novel residues; however, studies have shown that the COT gene can be activated as an oncogene by overexpression as well as by amino acid rearrangements.
COT has been shown to contribute to Epstein-Barr virus-encoded latent infection membrane protein 1-induced NF-kappaB signaling downstream of TRAF2.

Recommended Dilution:Immunohistochemistry (Formalin fixed paraffin embedded): 15ug/mlOptimal dilutions to be determined by the researcher.
Storage and Stability:May be stored at 4°C for short-term only.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Aliquots are stable for 12 months after receipt.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
