Maxi-K channels, also known as BK channels, consist of a pore-forming alpha subunit and a regulatory beta subunit, which confers the channel with a higher calcium sensitivity.
Homologs of the Drosophila melanogaster calcium-activated potassium channel gene, slowpoke, the mouse and human genes generate multiple transcripts by alternative splicing.
The splice variants differ significantly in their Ca2+ sensitivity, suggesting a broad functional role for these channels in the regulation of neuronal excitability.
The alpha-subunit shows a tissue distribution different to that of the beta-subunit, and in many tissues there may be no association of alpha-subunits with beta-subunits.
The beta-subunits can play a functional role in the regulation of neuronal excitability by tuning the Ca2+ and/or the voltage dependence of alpha-subunits (Tseng-Crank et al., 1996).
Maxi-K channels may have potential utility of gene therapy for urinary incontinence and erectile dysfunction.
