MKNK1 encodes a predicted 424aa protein containing a sequence typical of the catalytic domain of serine/threonine kinases and a putative N-terminal nuclear localization signal sequence.C-terminal region of MKNK1 was phosphorylated and activated in vivo and in vitro by ERK1 and p38 MAP kinases, but not by JNK/SAPK.
The activated ERK is essential for the activation of MKNK1 in insect cells.
MKNK1 was activated following stimulation of HeLa cells with a variety of extracellular stimuli.
MNK1 bind tightly to the growth factor-regulated MAP kinases, Erk1 and Erk2, and also binds strongly to the stress-activated kinase, p38.
Applications:Suitable for use in ELISA and Western Blot.
Other applications not tested.
Recommended Dilution:ELISA: 1:1,000Western Blot: 1:100-1:500Optimal dilutions to be determined by the researcher.
Storage and Stability:Store product at 4°C if to be used immediately within two weeks.
For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C.
Aliquots are stable at -20°C for 12 months after receipt.
Dilute required amount only prior to immediate use.
Further dilutions can be made in assay buffer.
Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Note: Applications are based on unconjugated antibody.
