• DNA-dependent protein kinases (DNA-PK) play a role in the repair of double-strand DNA breaks and in the process of V(D)J recombination during lymphoid development.
• By EST database searching for sequences homologous to the DNA-PK gene KIP/CIB and 5-prime RACE, Seki, N et al.
• isolated a full-length cDNA, which they designated as KIP2, from a human fetal brain cDNA library.
• KIP2 encodes a deduced 187aa protein with a predicted molecular mass of 22kD.
• The KIP2 protein shares 46%, 39%, and 30% sequence identity with the calcium-binding proteins KIP/CIB, calcineurin B and calmodulin respectively.
• KIP2 contains two EF-hand motifs and a helix-loop-helix motif involved in coordinating the calcium ion, indicating that KIP2 may also bind calcium.
• KIP2 is ubiquitously expressed in various human tissues.
• Applications:Suitable for use in ELISA, Western Blot, and Immunohistochemistry.
• Other applications not tested.
• Recommended Dilution:ELISA: 1:1,000Western Blot: 1:100-1:500Immunohistochemistry: 1:50-1:100Optimal dilutions to be determined by the researcher.
• Storage and Stability:Store product at 4°C if to be used immediately within two weeks.
• For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C.
• Aliquots are stable at -20°C for 12 months after receipt.
• Dilute required amount only prior to immediate use.
• Further dilutions can be made in assay buffer.
• Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates.
• For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
• Note: Applications are based on unconjugated antibody.
