Cyclin E1 belongs to the highly conserved cyclin family, whose members are characterized by a dramatic periodicity in protein abundance through the cell cycle.
Cyclins function as regulators of CDK kinases.
Different cyclins exhibit distinct expression and degradation patterns which contribute to the temporal coordination of each mitotic event.
Cyclin E1 forms a complex with and functions as a regulatory subunit of CDK2, whose activity is required for cell cycle G1/S transition.
Cyclin E1 accumulates at the G1-S phase boundary and is degraded as cells progress through S phase.
Overexpression of Cyclin E1 has been observed in many tumors, which results in chromosome instability, and thus may contribute to tumorigenesis.
This protein was found to associate with, and be involved in, the phosphorylation of NPAT protein (nuclear protein mapped to the ATM locus), which participates in cell-cycle regulated histone gene expression and plays a critical role in promoting cell-cycle progression in the absence of pRB.
Applications:Suitable for use in Immunohistochemistry, Flow Cytrometry and Western Blot.
Other applications have not been tested.
Recommended Dilution:Immunohistochemistry (formalin-fixed, parafin-embedded): 1:50-1:100Flow Cytrometry: 1:10-1:50Western Blot: 1:50-1:100Optimal dilutions to be determined by researcher.
Positive Control:A2058 cell line lysates, human testis tissue and NCI-H292 cells.
Storage and Stability:Store product at 4°C if to be used immediately within two weeks.
For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C.
Aliquots are stable at -20°C for 12 months after receipt.
Dilute required amount only prior to immediate use.
Further dilutions can be made in assay buffer.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Note: Applications are based on unconjugated antibody.
