Tribbles homolog 3 (TRB-3) is a pseudokinase, shown to act as a potent negative regulator of PPAR?, a master regulator of adipocyte differentiation, and to control adipogenesis.
It disrupts insulin signaling by binding directly to Akt kinases and blocking their activation.
It interacts also with the NF-kappaB transactivator p65 RELA.
May play a role in programmed neuronal cell death but does not appear to affect non-neuronal cells.
Applications:Suitable for use in Immunohistochemistry, ELISA and Western Blot.
Other applications not tested.
Recommended Dilution:ELISA (direct and indirect): 1:2000-1:5000Western Blot: 1:2000-1:5000 using ECL.
Suggested blocking and dilution buffer is PBST containing 0.05% Tween 20 and 5% skim milk.
Suggested incubation time is 1 hour at room temperatureImmunohistochemistry (paraffin sections): 1:500Optimal dilutions to be determined by researcher.
Storage and Stability:May be stored at 4°C for short-term only.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Aliquots are stable for 12 months after receipt.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
