Leptin is involved in the signaling pathway that regulates the size of the body fat depot.
An increase in the level of leptin may act directly or indirectly on the CNS to inhibit food intake and/or regulate energy expenditure as part of a homeostatic mechanism to maintain constancy of the adipose mass.
Defects in the leptin gene may be a cause of autosomal recessive obesity.
Applications:Suitable for use in ELISA and Western Blot.
Other applications not tested.
Recommended Dilution:ELISA (direct and indirect): 1:500-1:5000Western Blot: 1:500-1:5000 using ECL.
Suggested blocking and dilution buffer is TBST containing 0.05% Tween 20 and 5% skim milk.
Suggested incubation time is 1 hour at room temperature
Optimal dilutions to be determined by researcher.
Storage and Stability:Store product at 4°C if to be used immediately within two weeks.
For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C.
Aliquots are stable at -20°C for 12 months after receipt.
Dilute required amount only prior to immediate use.
Further dilutions can be made in assay buffer.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
