mRNA capping enzyme is one of the first mRNA transcription cycle factors to be associated with the C-terminal domain and functions by adding an inverted guanylyl group to the 5’ end.
mRNA capping enzyme also has triphosphatase and guanylyltransferase activity.
The CTD also serves to regulate the capping enzyme and has the ability to remove the Ser5 phosphate which is linked to the release of the capping enzyme from the capping complex.
mRNA capping enzyme has a variety of other functions during transcription including; stabilization of polymerase II, activation of early elongation and reduction of re-initiation.
