The p53 tumor suppressor is a transcription factor that functions to regulate cell proliferation and apoptosis.
The important of p53 in suppressing cancer is highlighted by the very high incidence of spontaneous tumor formation in p53-deficient mice and in mice bearing genetic alteration in other members of the p53 pathway.
P53 becomes stabilized and activated in response to inverse cellualar stressed such as DNA damage, hypoxia, ribonucleotide depletion and oncogene activation.
Phosphorylation occurs in response to DNA damage on human p53 serine residues 15 and 20 (mouse Ser18 and Ser23, respectively) and has been found in vitro studies to upregulate p53 activity.
Phosphorylation of serine 18 was required for robust p53-mediated apoptosis but not required for p53-mediated tumor suppression.
Phosphorylation at Ser20 (murine Ser23) of the p53 tumor suppressor is critical for disrupting p53 intraction with its negative regulator, MDM2, and allowing p53 stabilization.
Serine 23 phosphorylation in p53 plays an important role in p53 reponse to DNA damage in vivo and in tumor suppression.
