ATF6, a basic-leucine zipper protein, was isolated by yeast interaction assay as a binding partner to SRF (serum response factor).
SRF is a transcription factor, which binds to the serum response element (SRE) in the c-fos promoter and regulates expression of the c-fos gene as well as other immediate-early genes and some tissue-specific genes.
ATF6 has also been shown to interact with the endoplasmic reticulum (ER) stress response element (ERSE).
ATF6 constitutively expressed as a membrane-bound 90kD protein is directly converted to a 50kD protein in ER-stressed cells.
When unfolded proteins accumulate in the ER, ATF6 is cleaved to release its cytoplasmic domain, which enters the nucleus.
