Thymidylate synthase catalyzes the methylation of deoxyuridylate to deoxythymidylate using 5,10-methylenetetrahydrofolate (methylene-THF) as a cofactor.
This function maintains the dTMP (thymidine-5-prime monophosphate) pool critical for DNA replication and repair.
The enzyme has been of interest as a target for cancer chemotherapeutic agents.
It is considered to be the primary site of action for 5-fluorouracil, 5-fluoro-2-prime-deoxyuridine, and some folate analogs.
Expression of this gene and that of a naturally occuring antisense transcript rTSalpha (GeneID:55556) vary inversely when cell-growth progresses from late-log to plateau phase.
Applications:Suitable for use in Immunofluorescence, ELISA, Western Blot, Immunoprecipitation and Immunohistochemistry.
Other applications not tested.
Recommended Dilution:Immunohistochemistry (Formalin fixed paraffin embedded): 5ug/mlOptimal dilutions to be determined by the researcher.
Storage and Stability:Lyophilized powder may be stored at -20°C.
Stable for 12 months at -20°C.
Reconstitute with sterile dH2O.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Reconstituted product is stable for 12 months at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
