Interleukin-33 (IL-33), also known as NF-HEV and DVS 27, is a member of IL-1 cytokine family.
IL-33 is constitutively expressed in smooth muscle and airway epithelia and is upregulated by IL-1 Alpha or Beta in arterial smooth muscle, dermal fibroblasts, and keratinocytes.
IL-33 is synthesized as a 270aa, 32kD proform that localizes to the nucleus and may regulate gene transcription.
Cleavage generates mature IL-33 as an 18kD C-terminal proinflammatory fragment which binds IL-1 R4/ST2 on Th2 and mast cells.
Mature human IL-33 shares 55% aa identity with mouse IL-33.
