Secretory component is present in monkey secretions bound to secretory IgA (sIgA) and in free form.
Secretory IgA (sIgA) functions as a dimer or polymer and accounts for almost all specific mucosal antibody activity.
A molecule of sIgA is made up of two molecules of IgA, one J chain and one SC (MW 65000).
The dimer IgA is transported into secretions by its binding to the SC on the epithelial cells.
SC also has an affinity for polymeric IgM.
Applications:Suitable for use in ELISA, Immunohistochemistry, Western Blot, Immunocytochemistry, Immunoelectrophoresis and Double Radial Immunodiffusion.
Other applications not tested.
Recommended Dilution:ELISA: 1:200-1:4000Immunohistochemistry: 1:150-1:500Immunocytochemistry: 1:150-1:500Optimal dilutions to be determined by the researcher.
Storage and Stability:Lyophilized powder is stable for 12 months after receipt at -20°C.
Reconstitute with sterile ddH2O prior to use.
To avoid repeated freezing and thawing, aliquot reconstituted stock solution and store at -20°C.
Dilute required amount only prior to immediate use.
Further dilutions can be made in assay buffer.
Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

In immunoelectrophoresis against normalmonkey milk, using a high electroendosmosis agar plate, free secretory component is precipitated in the alpha-2 region. Des not react with other molecular forms of IgA, or with any other secretory or plasma protein and double radial immunodiffusion. May also react with free and bound secretory component in many old-world monkeys, including Macaca Sylvanus, Cercopithecus, Cynomolgus, Baboon and chimpanzees. In double radial immunodiffusion a reaction has been observed with free human secretory component.
