TYRO3 belongs to the “TAM” receptor family of receptor protein tyrosine kinases (RPTKs) composed of three receptors: TYRO3, Axl, and Mer.
TYRO3 is the primary TAM receptor detected in neurons of the central nervous system (CNS).
TYRO3 possess at its N-terminus two immunoglobulin-like domains (Ig domains) in the extracellular part, followed by two fibronectin-type III repeats (FNIII domains).
The membrane-spanning single helix is followed by the cytoplasmic tyrosine kinase domain.
TYRO3 serve as a potential modulator of postsynaptic neuronal excitability and also play a role in synaptic function in the central nervous system.
The protein is widely expressed in adult tissues, being most prominent in the brain, testis, lymphatic, kidney and vascular system.
The pathological role of the protein has been in variety of cancers including mammary tumors, leukemia, and non-small cell lung cancer cell lines.
Human TYRO3 gene is localized in the chromosomal region 15q15.1-q21.1.
