NALP10, a member of the NALP protein family, is reported to have a regulatory role in the innate immune system and forms a large signal-induced multiprotein complex, the inflammasome, which activates the pro-inflammatory caspases, caspase-1 and caspase-5.
Unlike other members of the family NALP10 lacks the LRR (leucine rich repeat) region but contains aDPIN domain and a NACHT domain.
Immunoprecipitation reports suggest that NALP10 interacted with ASC, CASP1, and IL-1B and inhibits NF-kB activation, CASP1/caspase-1-dependent IL-1B secretion and apoptosis induced by ASC (PYCARD) in a dose-dependent manner.
Expression of NALP10 is enhanced by stimulation with lipopolysaccharide.
Experiments with NALP10 suggest that it acts as a multifunctional negative regulator of inflammation and apoptosis.
Wide expression of NALP10 is reported with highest levels in heart, brain and skeletal muscle.
