Stearoyl-CoA desaturase (SCD, also known as Delta-9 desaturase, EC 1.14.99.5) is an iron containing membrane enzyme.
It catalyzes the rate limiting step in the biosynthesis of mono-unsaturated fatty acids.
The enzyme inserts a cis double bond between the C9 and the C10 positions in the acyl-CoA derivatives of saturated fatty acids such as palmitic acid and stearic acids to produce palmitoleoyl- and oleoyl-CoA, respectively.
Mono-unsaturated fatty acids play an important role in many processes, including energy metabolism (triacylglycerol storage in adipose tissues), anti- oxidation, lipid activated signal transduction, apoptosis, mitogenesis in certain tumors, and activate certain hormone receptors.
SCD genes are tightly regulated by signals such as insulin, leptin, carbohydrate, fatty acids and temperature.
A number of SCD genes have been identified (two SCD genes in rat, and SCD1 and SCD5 in human, and SCD1-4 in mouse).
All mouse SCD genes are located on chromosome 19.
The SCD-gene products are integral membrane proteins with four TM domains containing a novel sterol regulatory element binding protein (SREBP) binding site.
The SCD enzymes have a 30aa N-terminal sequence responsible for their rapid degradation and short half-life of 3-4 hours.
Most prominent expression of SCD1 is in lipogenic tissues (liver and adipose), SCD2 in brain, SCD3 in skin, preputial and harderian gland, and SCD4 in the heart.
SCD-4: The enzyme SCD4 (mouse 352aa, ~37-40kD; 79% identity with SCD1-3) expressed is restricted in the heat.
Liver X receptor alpha (LXR-alpha) agonists and high carbohydrate fat-free diet induces SCD4 expression.
Unlike SCD1, SCd4 expression is no repressed by dietary polyunsaturated fatty acids.
Leptin deficient ob/ob mice show elevated levels of SCD4 in the heart, and leptin treatment reduced SCD4 levels without affecting SCD1 and SCD2.
SCD1 deficient mice showed elevated levels of SCD4.
Applications:Suitable for use in ELISA.
Western Blot, though not tested, may potentially be used as an application.
Other applications not tested.
