IL-12 R-beta1 is a type I transmembrane protein belonging to the type I cytokine family of receptors.
It binds to IL-12 with low affinity.
The functional high-affinity IL-12 receptor is a complex of IL-12 R-beta1 and IL-12 R-beta2.
Applications:Suitable for use in Flow Cytometry.
Other applications not tested.
Recommended Dilutions:Flow Cytometry: 10ul labels 10e6 cells using D10.G4.1 mouse helper T cell lineOptimal dilutions to be determined by the researcher.
Flow Cytometry:This antibody has been tested for flow cytometry using D10.G4.1 cells
1.
Cells may be Fc-blocked with 1ug of mouse IgG/10e5 cells for 15 minutes at RT.
Do not wash excess blocking IgG from this reaction

After blocking, 10ul of I8434-49A was added to 1-2.5x10e5 cells and incubated for 30 minutes at RT

Unbound antibody was removed by washing the cells twice in F4568-10: Flow Cytometry Staining Buffer.
(1X).
Note that whole blood requires a RBC lysis step at this point using Flow Cytometry Mouse Lyse Buffer

The cells were resuspended in Flow Cytometry Staining Buffer for final flow cytometric analysis.
As a control for this analysis, cells in a separate tube should be treated with PE-labeled goat IgG antibody.
This procedure may need to be modified, depending upon the cell type and final utilization.
Individual users may need to titrate to determine the optimal reagent amount for their specific use.
Storage and Stability:Store product at 4°C in the dark.
DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid.
Dilute required amount only prior to immediate use.
Further dilutions can be made in assay buffer.
Caution: PE conjugates are sensitive to light.
For maximum recovery of product, centrifuge the original vial prior to removing the cap.
