MYB (myeloblastosis protooncogene) is an 80kD transcriptional activator.
It is found in hematopoietic and intestinal crypt epithelial cells.
MYB interacts with p300 and activates multiple genes, including Bcl2, GATA3, and MYC.
Although MYB is thought of as a pro-proliferation and antidifferentiation factor, it would now appear that MYB contributes to both T- and B-cell maturation and to cell survival in general.
Human MYB is 640aa in length.
It contains three DNA binding, HTH-myb type domains (aa35-193), one transcriptional activation segment (aa275-327), and a Leu-zipper motif (aa376-397).
There are multiple potential splice forms.
One shows an alternate start site at Met189 coupled with a 99aa substitution for aa567-640.
Another shows a 121aa insertion after Ser401.
Four show C-terminal substitutions: a 34aa substitution for aa314-640, a Met substitution for aa402-640, a seven aa substitution for aa317-640, and a 10aa substitution for aa603-640.
Over aa503-640, human MYB shares 84% aa identity with mouse MYB.
