This gene encodes the largest subunit of dynactin (DCTN), a macromolecular complex consisting of 10-11 subunits ranging in size from 22-150kD.
Dynactin binds to both microtubules and cytoplasmic dynein.
It is involved in a diverse array of cellular functions, including ER-to-Golgi transport, the centripetal movement of lysosomes and endosomes, spindle formation, chromosome movement, nuclear positioning, and axonogenesis.
This subunit interacts with dynein intermediate chain by its domains directly binding to dynein.
Alternative splicing of this gene results in at least 2 functionally distinct isoforms: a ubiquitously expressed one and a brain-specific one. Based on its cytogenetic location, this gene is considered as a candidate gene for limb-girdle muscular dystrophy.
Defects in DCTN1 are the cause of Perry syndrome (PERRYS) also called parkinsonism with alveolar hypoventilation and mental depression.
Perry syndrome is a neuropsychiatric disorder characterized by mental depression not responsive to antidepressant drugs or electroconvulsive therapy, sleep disturbances, exhaustion and marked weight loss.
Applications:Suitable for use in ELISA, Western Blot and Immunohistochemistry.
~150kD observed in cell l line A549 and in Human Testis lysates (MW 142kD)
Immunohistochemistry(Paraffin): 2-4ug/ml in paraffin embedded Human Cerebellum shows pixulate staining of cytoplasm in neuronal cells.
In paraffin embedded Human Testis shows cytoplasm staining in cells of the seminiferous tubules.
Optimal dilutions to be determined by the researcher.
Storage and Stability:May be stored at 4°C for short-term only.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Aliquots are stable for 12 months after receipt.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
