SMAD3 is a 50kD transcriptional modulator activated by TGF-beta and activin type 1 receptor kinase.
SMAD3 functions in cell proliferation, differentiation and development.
Upon stimulation, SMAD3 oligomerizes with SMAD4 and translocates into the nucleus where it binds to DNA and transcription factors.
SMAD3 activity induces differentiation of selected neuronal subtypes at the expense of other subtypes.
Studies have indicated that defects in SMAD3 may be a cause of colorectal cancer.
