Caspase-2 is required for the permeabilisation of the mitochondria and the resulting amplification of caspase activity, in response to cytotoxic stress.
Cytotoxic stress induced proteolytic cleavage of the caspase-2 proenzyme forms an active caspase which cleaves other caspases in a cascade.
This leads to the release of cytochrome c and Smac from mitochondria and the translocation of Bax from the cytoplasm to the mitochondria, leading to apoptosis.
In adults, caspase-2 is expressed in the placenta, lung, kidney, pancreas and at lower levels in the heart, brain, liver and skeletal muscle.
Applications:Suitable for use in Immunohistochemistry (paraffin) and Western Blot.
Other applications not tested.
Recommended Dilution:Immunohistochemistry (paraffin): 10ug/ml Western Blot: 1ug/ml Optimal dilutions to be determined by the researcher.
Storage and Stability:May be stored at 4°C for short-term only.
For long-term storage and to avoid repeated freezing and thawing, add sterile 40-50% glycerol, aliquot and store at -20°C.
Aliquots are stable for at least 12 months at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
