Human 5T4 (oncofetal antigen 5T4; also TPBG and trophoblast glycoprotein) is a 72kD glycoprotein member of the LRR family of proteins.
It is expressed on trophoblasts, tumor cells, ovarian cuboidal epithelium and embryonic stem cells, and impacts cell adhesion and motility.
The human 5T4 cDNA encodes a type I transmembrane protein precursor that is 420aa in length.
It contains a 324aa extracellular region (aa32-355) that shows one Ser-rich region followed by seven Leu-rich repeats (aa90-355).
Over aa31-355, human 5T4 shares 81% and 85% aa identity with mouse and canine 5T4, respectively.
Applications:Suitable for use in ELISA, Western Blot and Immunohistochemistry.
Other applications not tested.
Recommended Dilutions:Western Blot: 1ug/ml using HT-29 human colon adenocarcinoma cell line
Immunohistochemistry: 5-15ug/ml using immersion fixed paraffin-embedded sections of human placenta subjected to antigen retrieval reagent basic
Optimal dilutions to be determined by the researcher.
Storage and Stability:Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C.
Reconstitute with sterile PBS.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
