Human ADPN (Adiponutrin; also PNPLA3 and patatin-like phospholipase domain-containing protein 3) is a 53kD member of the PNPLA family of phospholipase A2 enzymes.
It is expressed in white adipocytes.
Adiponutrin is elevated in response to insulin and appears to act not as a phospholipase, but a molecule involved in anabolic pathways.
ADPN is likely to be a type II transmembrane protein that is 481aa in length.
It contains one patatin domain (aa10-179) that is associated with in vitro enzyme activity and a transmembrane segment between aa42-62.
There appear to be two splice variants.
One shows a four aa deletion of aa59-62 and a second shows an alternate start site at Met379.
Over aa 160-349, human ADPN shares 61% aa identity with mouse Adiponutrin.
Applications:Suitable for use in ELISA, Western Blot, and Immunohistochemistry.
Other applications not tested.
Recommended Dilution:ELISA: 0.5-1.0ug/mlWestern Blot: 1.0ug/mlImmunohistochemistry: 10ug/mlOptimal dilutions to be determined by the researcher.
Storage and Stability:Lyophilized powder may be stored at 4°C for short-term only.
Reconstitute to nominal volume by adding sterile 40-50% glycerol and store at -20°C.
Reconstituted product is stable for 12 months at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
