• Produced in goats immunized with purified, NS0-derived, recombinant human Punctin (rhPunctin; aa29-525).
• Human Punctin specific IgG was purified by human Punctin affinity chromatography.
• Punctin is a secreted glycoprotein resembling members of the ADAMTS family although it lacks the propeptide region and the metalloproteinase and disintegrin-like domains.
• Punctin is expressed in adult skeletal muscle and may have important functions in the extracellular matrix.
• Alternative splicing of the Punctin transcript results in three isoforms.
• Isoform 1 is the longest and contains the entire isoform 2.
• Isoform 3 has the same sequence as isoforms 1 and 2 until residue 361, but a unique C-terminal end of 78 amino acids.
• Applications:Suitable for use in ELISA, Western Blot, and Immunoprecipitation.
• Other applications not tested.
• Recommended Dilution:ELISA: 0.5-1.0ug/ml.
• The detection limit for rhPunctin is approximately 1ng/well.
• Western Blot: 0.1-0.2ug/ml.
• The detection limit for rhPunctin is approximately 2ng/lane and 5ng/lane under non-reducing and reducing conditions, respectively
• Immunoprecipitation: Used to immunoprecipitate rhPunctin from conditioned media of transfected NS0 cells.
• The recovered rhPunctin can be detected by western blot analysis using a monoclonal antibody
• Optimal dilutions to be determined by the researcher.
• Storage and Stability:Lyophilized powder may be stored at 4°C for short-term only.
• Reconstitute to nominal volume by adding sterile 40-50% glycerol and store at -20°C.
• Reconstituted product is stable for 12 months at -20°C.
• For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
• Further dilutions can be made in assay buffer.
