ADAM12 (A disintegrin and metalloprotease domain 12; also meltrin-alpha) is a 120kD type I transmembrane glycoprotein member of the metzincin family of proteases.
It is synthesized as a 909aa preproprecursor that contains a 25kD proregion (aa29-207) and a 92kD mature molecule.
Proteolytic cleavage in the Golgi removes the propeptide prior to cell-surface expression.
A 68kD soluble splice form exists in pregnancy where 36aa replace the C-terminal 205aa (705-909).
This circulating ADAM12 isoform remains associated with its prodomain via noncovalent linkage, possibly acting as an enzyme regulator.
Over aa29-513, human ADAM12 shares 78% and 79% aa identity with bovine and mouse ADAM12, respectively.
Applications:Suitable for use in Western Blot.
Other applications not tested.
Recommended Dilution:Western Blot: 0.1-0.2ug/ml Optimal dilutions to be determined by the researcher.
Storage and Stability:Lyophilized powder may be stored at -20°C.
Stable for 12 months at -20°C.
Reconstitute with sterile 40-50% glycerol, PBS..
Aliquot and store at -20°C.
Reconstituted product is stable for 6 months at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
